While fitness is important, skill is way more important in an MTB event than it is in a road race. And there is a tremendous skill difference between a novice MTB rider and an experienced rider
I entered a 40 mile (65 km MTB ) race a few years back and it took me a little over 4 hours to finish. I have never been so completely shattered at the end of a race - nothing else even comes close. I have done a lot of mountain biking but mostly on fast flowy single track. The race was on mountainous, twisty, rocky, rooty, beautiful, hellish trails. The apparent ease with which some riders handled the technical terrain was very impressive. I wasn't one of those riders.
Are you an experienced MTB rider? If you already have great MTB skills you can concentrate on building endurance for a long race.
If you haven't done much MTB riding I would try to do as much riding as possible on terrain that is similar to the 100 mile event. Try to hook up with local MTB groups and learn from better riders. Ride your MTB. A lot.
