Have bad coffee so that you CAN get the expt set up, with the understanding you will then reward yourself with GOOD coffee.

I've had most of it.  I have to be careful - if I have too much, my hands get shaky and that's not good for dealing with the tiny (0.2 mL) tubes.

That's so funny. I was just thinking of shaky hands and pipetting into gel wells this morning. Ruined many a lovely gel. Usually, of course, on the last well or two.

I've done that many times.  Usually I poke through them or tear the sides.  But I almost always have more wells than I need.

The worst is SDS-PAGE gels  that are vertical and have tiny openings for their well.  Makes agarose gels a piece of cake.

Ah yes I've spent many an afternoon bent over a bench loading up a denaturing PAGE gel and burning my hands pulling those little bitty tubes out of the heating block, fun times.

Aaaah, the memories. Sometimes I really miss this stuff. But I really can't spend a significant portion of my life in a lab or office--bad things happen, and not just a ruined gel or two.

HYPERPUPPY HYPERPUPPY HYPERPUPPY!

Although ... I'm beginning to think that everything people say about dogs is actually true of cats, too. Mine are stinky and slobbery and hyper and try to hump (well, nurse) my leg and they're very vocal and like to sniff and lick each other's butts.

I could never get coomassie to stain properly. Always ended up running duplicate gels and silver staining the second one after the coomassie didn't develop properly. Load the crap out of it and still the bands were weak.

Westerns: Whole nuther set of problems there. GAWD!

Jim, it could probably be a whole lot of things.  Could be any of the above, too much coffee, nerves, and I think some people are just shakey people.  Why do you ask?

Kido - 2010-10-05 1:24 PM If someone shakes while doing manual tasks, what could that be?

It sort of looked like they were straining, as if they had to apply a lot of force and shaking, but it was pretty easy...

MS?  Drinking problem?  There is no noticable shake day to day at the desk...

I'm just a shaky person.  Add to that too much caffeine and I am really bad.

Jim: Loading a gel is pretty tricky. You're trying to put a 2mm tip in a 5mm slot while holding the pipette about 9inches above where you're putting the tip. Plus looking through a layer of plastic and a tank full of water. Plus it's made out of gel, so go too deep or left or right too far and you've f'd it up. And the samples are warm and tips clog. PIA in lab work for sure.
